目的研究TNFa诱导蛋白3棚互作用蛋白1(TNFα-inducedprotein3-interactingprotein1,TNIPI)mRNA及其编码蛋白在进行期寻常型银屑病患者体内的表达水平,探讨其在银屑病发病机制中的意义。方法研究分为两组,病例组为2011年10月至2012年4月我院皮肤科收治的进行期寻常型银屑病患者25例,男性13例,女性12例,年龄(33.0±8.3)岁;健康对照组为我院体检中心健康人群共25例,男性14例,女性11例,年龄(31.3±9.9)岁。通过荧光定量PCR和Westernblot检测TNIPlmRNA及其编码蛋白ABIN1在病例组和对照组外周血淋巴细胞中的表达,免疫组化染色法检测ABINI蛋白在患者皮损和健康对照者皮肤中的定位和表达。结果病例组外周血淋巴细胞中TNIPImRNA的相对表达量(0.2921±0.1016)显著低于健康对照组(1.0859±0.2663)(P〈0.05),ABINl蛋白在病例组外周血淋巴细胞中的表达(0.0932±0.0039)与健康对照组(0.2909±0.0578)相比也有显著差异(P〈0.05)。ABINl蛋白主要表达在健康对照者皮肤的表皮层,且主要表达在细胞核外,病例组皮损中AB1N1蛋白的表达明显减少。结论银屑病患者TNIPImRNA及其蛋白的低表达可能与银屑病的发生、发展有密切的联系。
Objective To determine the difference of the expression of TNFeL-induced protein 3-inter- acting protein 1 (TNIP1) and its coding protein, A20-binding inhibitor of NF-KB activation (ABIN-1) between patients suffering from vulgaris psoriasis and health individuals in order to figure out the role of TNIP1 in the progress of vulgaris psoriasis. Methods Twenty-five psoriasis patients (13 males and 12 females, mean age of 33.0 ± 8.3 ) admitted to our department from October 2011 to April 2012 were enrolled. Twenty-five healthy control subjects (14 males and 11 females, mean age of 31.3 ±9.9) were subjected from the department of physical examination of our hospital. There was no significant difference between these 2 groups in age or gender. The mRNA expression of TNIPI and the expression of its translational protein ABIN1 in the peripheral lymphocyte were detected by QRT-PCR and Western blotting. Immunohistochemical assay was also employed to study the expression of ABIN1 in the skin. Results The expression of TNIPI mRNA in the peripheral lymphocyte was notably lower in vulgaris psoriasis group than the healthy control (0. 292 1 ±0. 101 6 vs 1. 0859 ±0.266 3, P 〈 0.05). The expression of ABIN1 in the peripheral lymphocyte was also remarkably lower in vulgaris psoriasis group(0.093 2 ±0.003 9)than the healthy group (0.290 9 ±0. 057 8, P 〈0.05). Besides, ABIN1 protein was over-expressed in the epidermis and mainly in the extranuclear in normal skin, but its expression level was very low in skin lesion of patients.
