以菜豆、蚕豆、黄豆的幼嫩叶片及其种子为材料,采用高盐低pH值法、SDS法和改良CTAB法3种不同的DNA提取方法提取其总DNA,用紫外分光光度法和琼脂糖凝胶电泳检测DNA质量。分析比较各种方法的提取效率,从中找出最适的DNA提取方法。结果表明,对于大豆、蚕豆、黄豆的幼嫩叶片,无论从得率上还是质量上看,高盐低pH值法均明显优于其它两种方法。
The total DNA was extracted from the bean, horse bean and soybean with methods of high salt, SDS and CTAB. The quality of the DNA was examined with spectrophotometer and agarose gel electrophocesis. Results showed that high salt was the optimum method in extracting the total DNA from three leguminous plants.
