目的研究磷脂酰肌醇-3激酶(PI3K)特异性抑制剂LY294002与姜黄素联合对人肾癌细胞的抑制作用。方法应用CCK-8法检测姜黄素单独或联合LY294002对人肾癌细胞786-O的抑制率;TUNEL及免疫荧光法检测caspase-3,观察姜黄素单独或联合LY294002对786-O细胞凋亡的影响。结果 LY294002联合姜黄素能显著增加姜黄素对786-O细胞的抑制率,同时显著增强姜黄素对细胞凋亡的诱导作用(P〈0.05)。结论 PI3K特异性抑制剂LY294002通过促进786-O细胞凋亡提高其对姜黄素的敏感性,抑制PI3K/Akt信号转导通路的活性能够提高姜黄素的肿瘤杀伤作用。
Objective To investigate the inhibitory effect of phosphatidylcholine-3 kinase(PI3K)specific inhibitor,LY294002,combined with curcumin on in vitro cultured renal carcinoma cells. Methods Renal carcinoma cells were treated with PI3 K specific inhibitor LY294002,curcumin alone,or in combination. CCK-8 assay was used to measure the inhibitory rate. TUNEL staining and immunofluorescence of caspase-3 were applied to evaluate the apoptotic effects. Results Inhibitory rate of 786-O cells was significantly increased under the combination administration of curcumin and LY294002,and the apoptosis were significantly induced(P〈0.05). Conclusion Sensitivity to curcumin of 786-O could be increased by LY294002 through the apoptosis-related mechanisms,and inhibition of PI3K/Akt pathway might be related with the activation of anti-tumor effect by curcumin.
