目的:采用响应曲面法对羌活-独活药对的超声辅助提取工艺进行优化。方法:在单因素试验基础上,采用HPLC法测定蛇床子素、异欧前胡素含量,并以蛇床子素、异欧前胡素得率的综合评分为指标,通过响应面法考察超声时间、甲醇体积分数、料液比、提取次数对羌活-独活混合提取工艺的影响。结果:响应面法超声提取羌活-独活药对中蛇床子素和异欧前胡素的最佳工艺参数:超声时间40 min,甲醇体积分数95%,料液比1∶15,提取次数为2次。在此条件下,蛇床子素和异欧前胡素的得率分别为(2.113±0.096)mg·g-1、(1.884±0.082)mg·g-1,均与理论预测值吻合。结论:该响应面法优选的提取工艺稳定可行,可用于药对羌活、独活中蛇床子素和异欧前胡素的提取。
Objective:Response surface methodology was used to optimize the extraction conditions of Notopterygii Rhizoma et Radix and Angelicae Pubescentis Radix. Methods:Based on one factor test,with composite score of extract amounts of osthole and isoimperatorin as index,response surface methodology was adopted to investigate effects of ultrasonic time,methanol volume fraction,material/liquid ratio and extraction times on extraction process. Results:The optimal extracting parameters of osthole and isoimperatorin from Notopterygii Rhizoma et Radix and Angelicae Pubescentis Radix by response surface methodology were as follows:ultrasonic time 40 min,methanol volume fraction 95%,material/liquid ratio 1∶15 and extraction twice. Under this condition,osthole and isoimperatorin were(2.113±0.096)mg·g~(-1)and(1.884±0.082)mg·g~(-1),respectively,which are in agreement with the theoretical predictions. Conclusion:This optimized extraction process was stable and feasible,it could be used to extract osthole and isoimperatorin.
