目的检测RECK蛋白及膜型基质金属蛋白酶MMP-14在神经母细胞瘤中的表达,探讨其在神经母细胞瘤的发生、发展、浸润和转移中的作用。方法选择临床及病理资料齐全的存档神经母细胞瘤蜡块标本36例,另取节细胞神经瘤蜡块标本10例作对照。采用第二代通用型二步法监测系统(PV-6000)免疫组织化学方法检测RECK及MMP14蛋白的表达。结果RECK蛋白在神经母细胞瘤组织中低表达(16.7%),并随肿瘤浸润深度的加深、远处转移的发生而降低(P=0.015〈0.05)。MMP-14在神经母细胞瘤组织中高表达(58.3%),并随肿瘤浸润深度的加深、远处转移的发生而增高(P=0.002〈0.05)。神经母细胞瘤组织中RECK与MMP-14蛋白的表达呈负相关(r=-0.418,P〈0.05)。结论神经母细胞瘤组织中RECK低表达,而MMP-14高表达。RECK及MMP-14可能成为评估神经母细胞瘤发生浸润转移的重要指标。
Objective To detect the expression of RECK (reversion-inducing cysteine-fich protein with Kazal motifs), MMP-14 (matrix metalloproteinase-14), as well as their correlations in neuroblastoma and to investigate their roles in metastasis and development of the tumor. Methods PV-6000 immunohistochemistry was used to measure the expression of RECK and MMP-14, in 36 samples of neuroblastoma tissue selected from paraffin wax-embodied specimens with complete clinicopathological data, and another 10 cases of ganglioneuroma tissue Results The expression rate of RECK in neuroblastoma was low (16. 7%), and reduced with the increased invasive depth and long-distance metastasis(P = 0. 015〈0. 05). The expression rate of MMP-14 in neuroblastoma was high (58. 3%), and elevated with the increased invasive depth and long distance metastasis (P = 0. 002%0. 05). The expression of RECK was negatively correlated with the expression of MMP-14 (r = - 0. 418, P〈0. 05). Concluions RECK is lowly expressed in neuroblastoma, but MMP-14 is highly expressed. RECK, MMP-14 may be served as the markers in the estimation of metastasis and dissemination of neuroblastoma.
