目的观察环维黄杨星D对正常人肝细胞(L-02)的体外细胞毒性作用。方法予以不同浓度的环维黄杨星D作用L-02细胞,MTT法检测细胞增殖情况;AnnexinV+PI双染色流式细胞术测定细胞凋亡率;倒置显微镜观察细胞形态;用酶标仪检测乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平以及ATP酶活力。结果环维黄杨星D可显著抑制L-O2细胞的增殖,改变细胞形态,促进细胞凋亡和LDH释放,降低Na+、K+-ATP酶、Ca2+、Mg2+-ATP酶及Ca2+-ATP酶活性;但对MDA和SOD无显著影响。结论一定剂量环维黄杨星D对肝细胞L-O2有损伤作用,其机制可能与抑制ATP酶活力有关。
Objective To explore in vitro cell toxicity of cyclovirobuxi- hum - D on normal human hepatocyte LO2. Methods LO2 were treated with different concentrations cyclovirobuxinum - D for 12,24,48 h. The proliferation was assessed by MTT. The rate of apoptosis was detected by using FCM with Annexin V -FITC and PI double labeling method. The changes of celluar morphology was investigated by inverted microscope. LDH, MDA, SOD contents and the activities of Na + , K + - ATPase, Ca2 + , Mg2 + - ATPase, Ca2 + - ATPase were determined by using enzyme mark instrument. Results Cyclovirobuxinum - D could significantly inhibit cell proliferation, cyclovirobuxinum - D changed celluar morphology. It increased significantly the rate of apoptosis and LDH level, decreased the activities of Na + , K + - ATPase, Ca2 + , Mg2 + - ATPase, Ca2 + -ATPase . There were no obvious changes in the SOD, MDA content. Conclusion A certain dose of cyclovirobuxinum -D could result in the cells damage effects. Mechanism of cyclovirobuxinum - D might be related with inhibition the activities of Na + , K + - ATPase, Ca2 + , Mg2 + - AT- Pase, Ca2 + - ATPase in cell membrane.
