Hemolin是昆虫的一种重要先天性免疫蛋白,属于免疫球蛋白超家族。克隆了家蚕hemolin基因,其序列全长5 100bp,含有5个外显子和4个内含子,基因cDNA编码含410个氨基酸的蛋白质,预测蛋白的分子质量约44.79 kD,pI 5.12。对该基因表达规律和生物学活性的结果研究表明:该基因只在家蚕蛹期脂肪体内特异性转录和表达,并能够被细菌和病毒诱导在幼虫期表达;该基因体外表达获得的重组蛋白具有抗菌生物学活性,能够有效抑制苏云金芽孢杆菌等的生长,由此暗示家蚕Hemolin蛋白可能在蚕体抵御病毒与细菌的感染过程中发挥重要作用。
Hemolin is an important insect innate immune protein belonging to the immunoglobulin superfamily.In this study,the hemolin gene in silkworm was cloned.It is 5100 bp long with 5 exons and 4 introns.The cDNA is 1 233 bp long and codes for a 410 amino acid protein with predicted molecular weight of 44.79 kD and pI of 5.12.Investigations on expression profile of the gene and biological activities of the expressed product showed that the silkworm hemolin gene was only expressed in fat body during pupal stage.Moreover,the expression could be induced by bacterial or viral infection during larval stage.The recombinant hemolin obtained from in vitro expression by using baculovirus expression system had antibacterial activity,being an effective inhibitor to the growth of Bacillus thuringiensis.These results imply that silkworm hemolin gene could play important roles in resisting infections from bacteria and viruses.
