目的探讨血管内皮生长因子(VEGF)和血管形成蛋白-1(Ang-1)对人源性心脏微血管内皮细胞(HCMECs)分泌血管形成蛋白-2(Ang-2)的影响。方法在正常或缺氧缺血清培养条件下,分别将100 ng/ml VEGF或Ang-1加入HCMECs中,孵育24 h,采用酶联免疫吸附法检测Ang-2水平。结果在正常培养条件下,VEGF处理组Ang-2水平高于空白组(P〈0.05),而Ang-1处理组Ang-2水平低于空白组(P〈0.05);在缺氧缺血清培养条件下,Ang-2分泌量显著性增加,VEGF处理组Ang-2水平高于缺氧缺血组、Ang-1处理组、空白组(P〈0.05)。结论缺氧缺血清和VEGF处理均提高HCMECs分泌Ang-2,Ang-1处理可以降低HCMECs分泌Ang-2。
Objective To study the effect of vascular endothelial growth factor(VEGF) and angiopoietin-1 (Ang-1) on secretion of angiopoietin-2 (Ang-2) in human cardiac microvascular endothelial cells(HCMECs). Methods On the condition of normal culture or hypoxic and deficient serum culture, 100 ng/ml of VEGF or Ang-1 cytokine was added into the HCMECs respectively, and the secretion level of Ang-2 was detected by enzyme-linked immunosorbent assay(ELISA) after the ?A-hour incubation. Results On normal culture condition,the Ang-2 level in the VEGF treatment group was significantly higher than that in the normal group ( P 〈 0.05 ), while the secretion level of Ang-2 in the Ang-1 treatment group was significantly lower than that in the normal group ( P 〈 0.05 ). On the condition of hypoxic and deficient serum culture, the secretion of Ang-2 increased significantly, the secretion level of Ang-2 in the VEGF treatment group was higher than that in the hypoxic and deficient serum group, Ang-1 treatment group and normal group ( P 〈 0.05 ). Conclusion Both hypoxic and deficient serum, and VEGF can enhance the secretion of Ang-2 in the HCMECs, while Ang-1 will decrease the secretion of Ang-2 in the HCMECs.
