目的探讨妊娠期恐伤刺激对其80日龄仔鼠情志和海马区去甲肾上腺素(NE)、肾上腺素(EP)的影响,并探讨两者的相关性。方法 2015年1月—2016年3月,选取80日龄SPF级Wistar大鼠59只(雌鼠34只,雄鼠25只),采用Morris水迷宫实验评价其认知水平。选取认知基线水平一致的30只雌鼠按照随机数字表法分为模型组和对照组,各15只,另选取认知基线水平一致且体格较强壮的15只雄鼠作为交配鼠。剩余雄鼠均作电击鼠用,剩余雌鼠备用代替不孕鼠。运用旁观电击法建立妊娠期恐伤大鼠动物模型。仔鼠延续母鼠分组,至80日龄时采用随机数字表法选取15只进行实验。采用旷场实验、糖水偏好实验、悬尾实验评价仔鼠情志;通过脑立体定位仪在右侧海马区采集脑透析液,应用高效液相色谱-电子俘获检测(HPLC-ECD)法检测NE和EP水平。结果模型组旷场实验水平得分、垂直得分,糖水偏好实验糖水消耗量、糖水偏好值均低于对照组,悬尾实验静止不动时间长于对照组(P〈0.05)。模型组各灌流时间点海马组织NE、EP水平均低于对照组(P〈0.05)。仔鼠海马组织NE水平与旷场实验水平得分、垂直得分以及糖水偏好实验糖水偏好值呈正相关,与悬尾实验静止不动时间呈负相关(P〈0.05)。仔鼠海马组织EP水平与旷场实验水平得分以及糖水偏好实验糖水偏好值呈正相关,与悬尾实验静止不动时间呈负相关(P〈0.05)。结论恐伤孕鼠可改变其80日龄仔鼠的情志及海马区NE和EP的水平,其情志改变与海马区NE和EP的水平密切相关。
Objective To investigate the status of emotion and norepinephrine( NE) and epinephrine( EP) in the hippocampus of 80-day-old neonatal rats of fear-impaired pregnant rats,and to study the correlation of emotional changes with NE and EP. Methods This study was conducted from January 2015 to March 2016. Fifty-nine 80-day-old SPF Wistar rats( 34 females and 25 males) were selected,and Morris water maze test was used to evaluate the cognitive level. Of them,30 females with the same cognitive level were selected and divided into the model group and control group by the random number table with 15 in each,15 strong males with the same cognitive level were selected as mating rats. The other male rats received electric shocks,while the other female rats were reserved to replace sterile rats. Fear-impaired pregnant rats were established by watching other rats being shocked by electricity. The neonatal rats were divided by the same method used by the fear-impaired pregnant rats and 15 of them were randomly selected for experiment on the 80 th day after birth. Emotional changes in them were observed by open field test,sucrose preference test and tail suspension test. Brain microdialysis of the right hippocampus was collected with the stereotaxis instrument to determine the level of NE and EP by HPLC-ECD. Results Compared with the control group,the scores of horizontal and vertical movements,consumption of sucrose and sucrose preference value of the 80-day-old rats in the model group were decreased,while the immobility time was increased( P〈0. 05); levels of NE and EP in the hippocampus of 80-day-old rats in the model group were decreased at different liquid perfusion time points( P〈0. 05). The level of NE in the hippocampus of the 80-day-old rats presented a positive correlation with the scores of horizontal and vertical movements,and sucrose preference value,while it showed a negative correlation with the immobility time( P〈0. 05).
