目的:探讨ADAM9、ADAM10基因沉默对腺样囊性癌高转移潜能细胞生物学行为的影响。方法:采用免疫组化检测15例腺样囊性癌原发灶和相应淋巴结转移组织中ADAM9、ADAM10蛋白的表达情况;采用RNA干扰方法抑制腺样囊性癌高转移潜能细胞系ACC-M、SACC-LM中ADAM9、ADAM10基因的表达,检测该基因沉默后对肿瘤细胞周期及体外增殖、转移能力的影响。采用SPSS11.0软件包对数据进行单因素方差分析及Fisher确切概率检验。结果:ADAM9、ADAM10蛋白在腺样囊性癌淋巴结转移组织中的表达比在原发灶组织中显著增高(P〈0.05);ADAM9、ADAM10基因沉默后细胞增殖能力下降(P〈0.05),肿瘤细胞出现S期阻滞;并且ADAM9、ADAM10基因沉默后,肿瘤细胞的体外侵袭能力显著下降(P〈0.01)。结论:ADAM9和ADAM10基因表达与腺样囊性癌的转移有关。ADAM9、ADAM10基因沉默可改变腺样囊性癌细胞体外增殖能力、侵袭能力等生物学特性。
PURPOSE: To evaluate the effects of ADAM9 and ADAM10 gene silence on biological behaviors of high metastatic potential human adenoid cystic carcinoma cells. METHODS: The expression of ADAM9 and ADAM10 protein was examined both in 15 primary human adenoid cystic carcinoma tumors and their corresponding metastatic lymph nodes. ADAM9 and ADAM10 genes were suppressed in ACC-M and SACC-LM cell lines by RNA interference, and the cell growth capacity, cell cycle changes and metastatic capacity were then detected.One-way ANOVA and Fisher's exact test were used for statistical analysis with SPSS11.0 software package. RESULTS: The expression of ADAM9 and ADAM10 was significantly higher in metastatic lymph nodes metastasis than in primary tumors(P〈0.05). The ADAM9 and ADAM10 genes were effectively silenced by RNA interference. An Inhibition of cell proliferation and S cell-cycle arrest was observed in these treated cells. Moreover, the metastatic capacity of the cells in vitro was significantly decreased(P〈0.01). CONCLUSIONS: It is suggested that the ADAM9 and ADAM10 genes are associated with the metastasis of adenoid cystic carcinoma. Down-regulation of these genes could aher the biological behaviors of human adenoid cystic carcinoma cells.
