T-bet和Gata-3基因在脐带血中的表达特点

目的:了解转录因子T-bet和Gata-3基因在脐带血中的表达情况。方法:采用实时定量RT-PCR检测21例脐带血单个核细胞(CBMC)中转录因子T-bet和Gata-3基因的表达水平,21例健康成人外周血单个核细胞(PBMC)作为对照。结果:T-bet基因和Gata-3基因在CBMC和PBMC中均存在一定表达,Gata-3基因在CBMC中的表达水平(0.43±0.35)%与PBMC(0.42±0.27)%相比无明显差异(P=0.925),而T-bet基因在CBMC中的表达水平(0.47±0.48)%则明显低于健康成人组(2.12±1.48)%(P=0.000)。结论:CBMC中T-bet基因表达低下,可能与脐带血移植后GVHD发生率低下以及发生程度较轻有关。

英文摘要:

Aim:In order to evaluate the expression level of transcription factors T-bet and Gata-3 mRNA in umbilical cord blood from humans. Methods: Real-time PCR was used for detecting T-bet and Gata-3 genes expression level in 21 cases of mononuclear cells from umbilical cord blood(CBMC).21 cases of peripheral blood mononuclear cells(PBMC) of healthy adults served as controls.Results: T-bet and Gata-3 genes were expressed in all cases from both CBMC and PBMC.The expression level of Gata-3 gene(0.43%±0.35%) in CBMC was not significant difference in comparison with PBMC(0.42%±0.27%)(P=0.925).Compared with PBMC(2.12%±1.48%),the expression level of T-bet gene from CBMC was obviously down-regulated(P=0.000).Conclusion:The down-regulated expression of T-bet gene might relate to the reduced incidence and severity of GvHD after allogeneic transplantation of umbilical cord blood.
