丝裂原活化蛋白激酶(MAPK)信号通路是生物体内重要的信号转导系统之一,其亚族主要包括细胞外信号调节激酶1/2(ERK1/2)、c-Jun氨基末端激酶(JNK)、p38丝裂原活化蛋白激酶(p38MAPK)和细胞外信号调节激酶5(ERK5)。近年来研究发现,ERK5对细胞的增殖、分化及调节细胞功能有重要作用,并参与一些疾病的发生和发展,故阐明ERK5通路的作用机制,将为一些疾病的诊治提供新的思路与方法。现重点对ERK5的蛋白结构、生物学功能以及与疾病的关系进行综述。
Mitogen-activated protein kinase (MAPK) signaling pathway is one of the important in vivo signal transduction systems. The subfamily includes ERK1/2, JNK, p38 and ERKS. Recently studies found that ERK5 plays an important role in cell proliferation, differentiation and regulation of cell functions, in- volved in occurrence and development of some diseases. Therefore, the clarification of mechanism of ERK5 pathway will provide new thoughts and methods for diagnosis and treatment of the diseases. Here is to make a review on the current ERK5 research on protein structure, biological function and the relationship with the diseases.
