目的探讨在乳头状甲状腺癌K1及MCF7细胞系中,雌激素(E2)对hTERT的mRNA及蛋白表达的影响。方法应用Western Blot检测乳头状甲状腺癌K1细胞中雌激素受体(ER)的表达;应用共聚焦激光扫描显微镜观察ER在E2刺激后定位的改变;应用Real-time PCR检测E2刺激对人端粒酶逆转录酶(hTERT)mRNA表达的影响;应用Western Blot检测E2刺激对hTERT的蛋白表达的影响。结果 K1细胞系中存在ERα阳性表达,在E2刺激后阳性表达产物入核,E2刺激能够促进K1细胞中hT ERTmRNA和蛋白表达水平上调。结论雌激素促进hT ERT在乳头状甲状腺癌细胞中的表达。
Objective To investigate whether estrogen modulates the expression of human telomerase reverse transcriptase(hTERT) in papillary thyroid cancer cells. Methods The expression of ER α in K1 cells was detected by Western blot, the localization of ER α by E2 stimulation was observed using laser scanning confoeal microscopy, the expression of hTERT mRNA and protein by E2 stimulation was detected by Real-time PCR and Western blot respectively. Results ERa-positive cells was found in K1 cell line, and ERα positive product was translocated into nucleus after E2 stimulation. The expression levels of hTERT mRNA and protein were increased after E2 stimulation. Conclusion Estrogen upregultes hTERT expression in papillary thyroid cancer K1 cells.
