分别采用硫酸铵盐析和蔗糖密度梯度离心结合离子交换层析方法对紫细菌中的固氮红细菌(Rhodobacter azotoformans)色素蛋白复合体进行分离纯化,获得了不同光谱类型的色素蛋白复合体,并分析了色素蛋白复合体多样性原因。蔗糖密度梯度离心结合离子交换层析法分离到光反应中心和1种外周捕光复合体;硫酸铵盐析结合离子交换层析法分离到光反应中心、3种具有不同A798/A848比值的外周捕光复合体,还分离到含量较低的具有423nm异常吸收峰的外周捕光复合体。由此可见,在常规培养条件下,硫酸铵盐析结合离子交换层析能纯化获得多种光谱类型的色素蛋白复合体。
Using ammonium sulfate fractionation and sucrose densitygradient centrifugation combined ion exchange chromatography to isolate and purify pigment protein complex from Rhodobacter azotoformans R7.The different types of pigment protein complexes were obtained and the origin of the spectral difference was discussed.The results showed that the sucrose densitygradient centrifugation combined ion exchange chromatography yielded reaction center and one type of peripheral light harvesting complex;while ammonium sulfate fractionation combined ion exchange chromatography yielded reaction center,3different spectral types of peripheral light harvesting complexes and a further type of peripheral light harvesting complex with new spectral feather.In conclusion,ammonium sulfate fractionation combined ion exchange chromatography can yield multiple spectral types of pigment protein complexes.The present study provides methods for obtaining different spectral types of light harvesting complex 2.
