受体激酶是一类具有信号肽序列、胞外受体结构区、跨膜区和保守的胞内激酶区的蛋白质,普遍存在于各种生物体内.植物类受体激酶是植物体内信号分子的重要受体,几乎参与了所有的细胞代谢过程,包括生长发育、表达调控和逆境反应等.鉴于激酶的重要性和特殊性,其底物筛选已经成为该领域的研究热点.目前,筛选植物激酶底物的方法主要有酵母双杂交技术和蛋白质芯片技术等,但是,已经进行体内鉴定的底物极其有限.本文总结了植物类受体激酶的功能及其底物与筛选鉴定方法,并介绍了一些激酶的底物.
Receptor kinase is widespread in various organisms, and the majority of them are composed of a signal peptide, an extracellular domain, a single transmembrane and a cytoplasmic region which contains a conserved kinase domain. Plant receptor-like kinases are important sensors in various signaling pathways and play critical roles in a variety of biological process, including development and cell growth, gene expression regulation, and disease resistance, et al. The substrate screening and identification of these kinases has become a hot area in the researches to reveal their functional importance and the specialization. Yeast two- hybridization and protein microarrays are currently the primary approaches for identifying protein kinase substrates in vitro, and a couple of the candidates have been verified in vivo. The methodology for kinase substrate screening and identification, the properties of the substrates, and the functions of the kinases will be discussed in this review.
