翻译控制肿瘤蛋白(translationally controlled tumor protein,TCTP)是一种在生物体中普遍存在和结构上高度保守的多功能蛋白。本研究以橡胶树HbTCTP1基因的开放阅读框(open reading frame,ORF)作为诱饵,采用酵母双杂交系统构建筛选HbTCTP1互作蛋白的诱饵表达载体pBD.GALA—HbTCTP1,并在pADGAL4—2.1质粒构建的橡胶树胶乳cDNA文库中筛选HbTCTP1互作蛋白。将获得的两个与HbTCTP1互作的阳性克隆进行测序,比对NCBI非丰富蛋白数据库发现:它们分别是橡胶延长因子(rubber elongation factor,REF)和60S核糖体L44蛋白(60S ribosomal protein L44,RPL44)。本研究结果对进一步揭示HbTCTP1在橡胶树中的作用具有重要意义。
Translationally controlled tumor protein is a multifunctional protein that is highly conserved and widely present in organisms. In this study, the proteins interacting with HbTCTP1 were screened by yeast two-hybrid system. A bait vector pBD-GAL4-HbTCTP1 was constructed by subcloning Hb TCTP10RF into yeast two-hybrid vector pBD-GAL4 Cam, and pAD-GAL4-2.1 cDNA prey library of rubber tree latex was prepared. The yeast strain YRG-2 was sequentially transformed with the bait pBD-GAL4-HbTCTP1 and latex cDNA library plasmid DNA by the lithium acetate method. Two positive clones interacting with HbTCTP1 were sequenced and searched against NCBI non-redtmdant protein database. The results indicated that the two interaction proteins were rubber elongation factor (REF) and 60S ribosomal protein RPL44. The results are of great significance for further studying Hb TCTP1 roles in rubber tree.
