使用辛酸沉藏-凝胶过滤法提取猪血清中免疫球蛋白IgM,并通过其纯度鉴定等探讨提纯效果。通过辛酸沉淀法粗提免疫球蛋白,再结合Sephadex G200凝胶过滤法纯化获得IgM,利用SDS—PAGE电泳法和AlphaEaseFC凝胶成像分析软件测定其分予量和纯度、Bradford法测定提取蛋白浓度并计算其得率.结果显示辛酸粗提能够去除大部分杂蛋白,凝胶过滤获得两个洗脱峰,其中第一峰通过电泳鉴定含有IgM,纯度63.8%,得率1.28mg/ml血清.利用辛酸沉淀-凝胶过滤法能够获取纯度和得率都较高的IgM,纯度可满足普通要求的实验和应用。
Purify the immunoglobulin M (IgM) from swine serum with the caprylic acid precipitation-the Sepixerdax G200 gelatin filters and approach the purifying effect by identifying its purity. The experiment combines the eaprylic acid precipitation method and the Sepherdax G200 gelatin filters to purify IgM. Then the molecular weight and purity of IgM are de termined by SDS-PAGE and AlphaEaseFC analysis software, and the yield of IgM is calculated according to the protein concentration determined by Bradford kit. The caprylie acid precipitation can eliminate the bulk of proteins which are not IgM. Two eluting peaks are abtained in the process of purification IgM by the Sepherdax G200 gelatin filters. The purity of IgM which is only in the first eluting peak is 63.8%; and the yield is 1.28 mg/ml serum. The experiment can get high pnrirv. and yield of IgM by the caprylic acid precipitation method and the Sepherdax G200 gelatin filters. The IgM purified horn swine serum in this experiment can meet the need of the experiments and the applications,in which the requirement or purity is, net high.
