对产苦马豆素菌株Aspergillus ustus发酵条件进行优化,采用正交试验方法对发酵过程中的营养因子(碳源、氮源)和环境因子(基础培养时间、温度、装样量、pH等因素)进行筛选,运用气相色谱仪对菌株Aspergillus ustus不同培养条件下的苦马豆素产量进行检测。结果显示,产苦马豆素菌株Aspergillus ustus的最适发酵条件是豆粕培养基,硝酸钾4mg,麦芽糖20mg,装样量250mL,pH 6.0,温度35℃,发酵时间4周。本试验为苦马豆素来源提供了新途径,为苦马豆素工业化生产奠定了基础。
In order to explore the effect of different nutrition factors and culture conditions on the swainsonine production by microbial fermentation,the study preliminarily optimized the fermenta- tion conditions of Aspergillus ustus, screening the nutrition factors (culture medium, carbon source, nitrogen source) and the environment factors (cultivation time, temperature, fermentation volume and pH) by orthogonal design experiment and detecting the swainsonine production of Aspergillus ustus in different culture cconditions by GC. The results showed that the best fer- mentation condition of Aspergillus ustus was soybean meal medium,4 mg potassium nitrate as ni- trogen source,20 mg maltose as carbon source,250 mL volume in 500 mL conical flask,pH 6.0, temperature 35 ℃ and 4 weeks fermentation. The study provided a new way for swainsonine source and laid the foundation for swainsonine industrial production.
