目的:观察12-去氧佛波醇13-棕榈酸酯(12-Deoxyphorbol-13-Palmitate,12D-13P)对慢性粒细胞白血病(chronic myeloid leukemia,CML)细胞株K562细胞增殖的抑制作用.方法:不同浓度的12D-13P处理的K562细胞,光镜下观察细胞形态学变化,MTT法检测药物对K562细胞增殖的抑制作用,甲基纤维素克隆形成实验分析细胞克隆能力.结果:MTT法和甲基纤维素克隆形成实验均显示12D-13P对K562细胞增殖具有较明显的抑制作用.结论:12D-13P能抑制体外培养的K562细胞的增殖,且呈一定的量效和时效关系.
Objective:To observe the inhibitive effect of 12-Deoxyphorbol-13-Palmitate(12D-13P)on the cell proliferation of chronic myeloid leukemia cell line K562.Methods:K562 cells were treated with different concentrations of 12D-13P.Morphology changes were observed with optical microscope.Medicine role in cell proliferation was detected with MTT assay.Cell cloning ability was analyzed with methyl cellulose colony-forming experiment.Results:MTT methods and methyl cellulose colony-forming experiment indicated 12D-13P had an obviously inhibitory effect on K562 cells.Conclusion:12D-13P can significantly inhibit the growth of in vitro cultured K562 cells and it shows a dose effect and time effect relationship.
