毒作用模式(modeofaction,MOA)在环境化学物危险度评价中发挥了极其重要的作用。近年来,生物标志的研究进展和组学技术(0mics)在毒理学研究中的应用为改进MOA研究、完善其在人类健康危险度评价中的应用提供了新思路。该综述主要介绍了近年来MOA在危险度评价中的应用进展,以MOA的建立及人类关联性框架为重点,以黄曲霉毒素B1(aflatoxinB1,AFB1)为例阐述MOA的研究思路和分析方法。此外,还介绍了Omics技术在MOA研究中的应用趋势及MOA在环境化学物危险度评价中新的应用取向——替代终点。
Mode of actions (MOAs) is of great importan in risk assessment of environmental chemical pollutants. Recently, great progresses have been made in terms of omics and biomarkers, which lead to development of MOAs studies. These new technologies and methodologies are expected to make better use of MOAs in health risk assessment. Here, the progress in definition and constitution of MOA was reviewed, in relation to human relevance framework (HRF) and case analysis illustrated by aflatoxin Bf. The application of omics technology in MOAs study and the potential usage of surrogate endpoint in risk assessment were also presented.
