目的探讨帕金森病(PD)伴发急、慢性抑郁大鼠模型的行为学及其不同脑区中单胺类神经递质的改变。方法成年雌性SD大鼠随机分为正常组、PD组、PD+慢性抑郁组、慢性抑郁组、PD+急性抑郁组、急性抑郁组。在6一羟基多巴胺(6-hydroxydopamine,6-OHDA)所造的PD大鼠模型的基础上建立慢性抑郁或急性抑郁大鼠模型。于不同的时间点,测定体重的变化,采用旷场实验、被动回避实验评价大鼠抑郁程度及学习能力,观察各组大鼠模型不同脑区(额叶、黑质、海马、蓝斑)单胺类神经递质水平的变化。结果①PD组及慢性抑郁组与正常组相比在第7、14、21天体重增长率明显降低,PD+慢性抑郁组与其他各组相比,上述各个时间点体重增长更慢,差异有统计学意义。②旷场实验中,PD+慢性抑郁组各时间点水平运动总距离及垂直得分与其他各组相比均低,尤其在第1天及第7天。急性抑郁组及PD+急性抑郁组在第1天、第3天水平运动总距离及垂直得分亦有显著变化。③被动回避测试中,与正常组相比较,PD组和慢性抑郁组从第7天开始,T1值随时间延长而逐渐增加,T2值随时间延长而逐渐减少,差异有统计学意义。而相比于上述3组,PD+慢性抑郁组从第1天开始,T1、T2值变化更为显著。④在各脑区,PD组、慢性抑郁组、急性抑郁组之间5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)水平差异无统计学意义,均较正常组显著减少;而PD+急性抑郁组、PD+慢性抑郁组5-HT、NE及DA水平较上述组减少更加显著。结论PD伴发抑郁的机制为内源性因素所致,而外源性因素起了促进及加重作用。
Objective To explore the behaviors of models of 6-hydroxydopamine (6-OHDA)-induced Parkinson's disease (PD) with acute or chronic depression and the changes of monoamine neurotransmitters in different brain regions. Methods Fe- male adult rats were randomly divided into normal group, PD group, PD with chronic depression group, chronic depression group,PD with acute depression group, and acute depression group. The chronic depression model and the acute depression model were established based on the model of 6-OHDA-induced PD. The body weight of the rats was dynamically measured at different time. The open-field test (OFT) and the passive avoidance test were used to evaluate depression-like behavior and the ability of learning. Enzyme-linked immunosorbent assay was used to measure the contents of 5-HT,NE,and DA in the brains of the rats. Results @The growing rate of the body weight of the rats was significantly decreased in PD group and chronic de- pression group compared with the normal group at 7,14 and 21 days. The growing rate of the body weight in PD with chronic depression group was lowest at all measured time points. QOFT showed that the total horizontal distance and the vertical move- ment score were significantly decreased in the PD with chronic depression group when compared with those in other groups,es- pecially at 1 day and at 7 days. Also,they were significantly reduced in the acute depression group and PD with acute depression group at 1 day and at 3 days. @The passive avoidance test showed that the T1 value was significantly increased and the T2 val- ue was significantly decreased in a time-dependent manner in PD group and chronic depression group compared with the normal group from the 7th day. The changes of the T1 value and T2 value were more significant in PD with chronic depression group compared with the other three groups from the 1st day. @ There were no significant differences in the contents of 5-HT, NEand DA in different brain regions among PD group,chronic depression group
