利用高效液相色谱法对肉鸭肝脏中参与脂肪酸合成关键酶——脂肪酸合成酶的活性进行分析,并测定了鱼腥草素对鸭肝脂肪酸合成酶活性的影响。采用离心法从鸭肝中分离纯化脂肪酸合成酶,并用鱼腥草素(100 mg/L)处理脂肪酸合成酶,然后用HPLC法检测还原型NADPH在340 nm处吸收峰变化。HPLC最佳测定条件为:Hypersil C18色谱柱,流动相:0.01 mol/L磷酸盐缓冲液(pH7.2)-甲醇溶液(80∶20,V/V),柱温:25℃,流速:1 mL/min,检测波长:340 nm,进样量:20μL。HPLC加样的回归方程:A=17754x+123.3,r=0.9945。实验结果表明,还原型烟酰胺腺嘌呤二核苷酸(NADPH)浓度在0.5μmol/L~1.44 mmol/L内线性关系良好。HPLC法检测鸭肝脂肪酸合成酶的活性,样品用量少,准确度高。
A high performance liquid chromatography method measuring the fatty acid synthase (FAS) activity and the effect of HOU-Cn to FAS activity were established. FAS from duck liver was treated with houttuyfonate homologues ( HOU-Cn ). Then the change in the activity of FAS was determined by HPLC. The optimal conditions of HPLC were as follows: Hypersil C18 column, mobile phase: 0.01 mol/L phosphate buffer (pH 7.2)- methanol (80:20, v/v), column temperature: 25 ℃, flow rate: 1 mL/min, detection wavelength: 340 nm, injection volume : 20 μL. The regression equation of HPLC was : A = 17754x + 123.3, r = 0.9945. An excellent linearity over the range of 0.5 μmol/L - 1.44 mmol/L was observed. The HPLC method can be used to determine the FAS activity with less dosage and high accuracy.
