目的:研究灵芝三萜酸(GTA)对四氯化碳(CCl4)和白酒所致小鼠急性肝损伤的保护作用。方法:采用CCl4和56度白酒分别制备小鼠急性肝损伤模型。赖氏法检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的水平;硫代巴比妥酸法测肝匀浆丙二醛(MDA)含量;黄嘌呤氧化法测肝匀浆超氧化物歧化酶(SOD)活力;HE染色观察肝脏病理形态学改变。结果:GAT能降低急性肝损伤小鼠血清中ALT、AST活性,降低肝匀浆MDA含量,提高肝匀浆SOD活性,并不同程度减轻动物肝脏损伤的程度。结论:灵芝三萜酸对急性肝损伤有一定的保护作用,其机制可能与其抗脂质过氧化作用有关。
OBJECTIVE To investigate the protective effect of Ganoderma tritepenoidie acid (GTA) on acute liver injury in mice. METHODS The experimental models of acute liver injury were induced by carbon tetrachloride(CC14 ) and 56 degree liqueur in mice. The activities of ALT, AST in serum were determined with Reitman-Frankel method, MDA content and SOD activity in liver homogenate were measured with thiobarbituric acid method and xanthine oxidase method, respectively. The liv er pathological morphological changes were detected by HE staining. RESULTS GTA could reduce the activities of ALT, AST in serum and the contents of MDA in liver homogenate of acute liver injury mice models. It could raise the activity of SOD in liver homogenate. GTA improved the histopathologic change of liver injury in different degrees. CONCLUSION GTA showed certain protective effect against acute liver injury and the mechanism may be related to its anti-lipid peroxidation.
