目的:评价新型免疫抑制剂来氟米特应用对CD4^+CD25^+调节性T淋巴细胞的影响。方法:以灌胃的方式给予正常BALB/c小鼠来氟米特或对照溶剂4周,流式细胞仪分析不同淋巴器官内T淋巴细胞亚群,包括CD4^+CD25^+调节性T细胞数量及比例的变化。结果:与对照溶剂组相比,LEF组小鼠脾、肠系膜淋巴结内CD4^+T淋巴细胞和CD4^+CD25^+调节性T细胞比例均明显下降,外周淋巴结和胸腺没有明显变化;但胸腺内CD4^+CD25^+调节性T细胞/CD4单阳性胸腺细胞的比例明显增加。结论:来氟米特对中枢和周围淋巴器官内CD4^+CD25^+调节性T细胞的影响不同。胸腺内CD4^+CD25^+调节性T细胞对来氟米特的作用与CD4单阳性胸腺细胞相比具有较强的抗性;而脾及淋巴结的CD4^+CD25^+调节性T细胞对来氟米特的反应与非调节性T细胞相似。
Objective:To evaluate the effect of leflunomide(LEF)on CD4^+CD25^+ regulatory T cells(Treg).Methods:BALB/c mice were given LEF or vehicle solution by gastric gavage for 4 weeks.The levels of different T subsets,including CD4^+CD25^+Treg cells in spleen,lymph nodes and thymus in both groups were then detected by flow cytometry(FCM).Results:The percentages of CD4^+ T cells and CD4^+CD25^+Treg cells from spleen and mesenteric lymph nodes were significantly reduced after the treatment with LEF(P〈0.05),but no differences were detected in peripheral lymph nodes(axillary,brachial,inguinal) and thymus.The total cell number and percentage of CD4+CD8-thymocytes in the thymus decreased,but the total number of CD4^+CD25^+Treg cells did not alter,which leaded to the significant increase in the ratio of CD4^+CD25^+Treg cells to CD4^+ T cells in the thymus.Conclusion:Our data indicate that LEF has different effects on CD4^+CD25^+Treg cells from central and peripheral lymph organs,and the thymic CD4^+CD25^+Treg cells could be resistant to the effects of LEF,while CD4^+CD25^+Treg cells in peripheral immune organs have the same responses to LEF as non-regulatory T cells.
