目的:观察中药验方痒咳宁干预喉源性咳嗽模型时Th/Treg相关特异性炎性因子的变化,评价中药的疗效。方法:以多因素复合模拟中医病因建立喉源性咳嗽模型,用Real-time PCR技术检测中药痒咳宁干预模型豚鼠肺组织转化生长因子-β(TGF-β)、叉头状转录因子p3(Foxp3)、白细胞介素-10(IL-10)、白细胞介素-17(IL-17)m RNA的表达。结果:与正常组比较,模型组豚鼠出现体质量减轻、咳嗽次数增加,咽喉、肺组织病理组织均显示存在不同程度的炎性反应,血中的炎性细胞尤其是嗜酸粒细胞比例等出现升高(P〈0.05),Th/Treg相关特异性炎性因子TGF-β、Foxp3、IL-10、IL-17的表达出现异常。用药后,中药各组豚鼠出现体质量增加(P〈0.05),咽喉、肺组织病理组织的炎性反应逐渐好转,各特异性炎性因子出现不同程度的缓解,其中中药中、高组对Th/Treg炎性通路有明显干预作用(P〈0.05)。结论:验方痒咳宁疗效确切,可能通过影响Th/Treg相关特异性炎性因子起效。
Objective: To evaluate the effects of Yangkening Formula on special inflammatory factors related to Th/Treg in guineapig with laryngeal cough. Methods: The laryngeal cough model was established by various factors that imitated the pathogenic factors of traditional Chinese medicine. The real-time PCR technology was used to detect the expressions of TGF-β, Foxp3, IL-10 and IL-17 m RNA in the lung tissue of guineapig. Results: After modeling, the guineapigs showed the body weight loss, an increase of cough, inflammatory reaction in lung tissue and an increase of eosinophils granulocyte in blood. In addition, special inflammatory factors related to Th/Treg showed abnormal expressions, including TGF-β, Foxp3, IL-10 and IL-17. After treatment, the guineapigs showed an increase of the body weight(P0.05), a decline of cough and the improvement on inflammatory reaction and inflammatory factors. What's important was that the lymphocyte was significantly intervened by Yangkening Formula at a moderate dose and a high dose(P0.05). Conclusion: Yangkening Formula could effectively improve the laryngeal cough, which might be related to influence in the special inflammatory factors related to Th/Treg.
