以广西钦州产花生壳为原料,用乙醇回流提取法提取花生壳中的黄酮物质,采用分光光度法测定提取物中的黄酮含量。试验以提取量为指标,在不同温度、时间、液料比、乙醇浓度的工艺条件下提取花生壳中的黄酮物质,并采用L9(34)正交实验设计,对提取工艺条件进行优化。通过分析确定出最佳的提取工艺条件为:提取时间2.5 h,乙醇浓度70%,液料比30∶1,提取温度80°C,在此条件下得到黄酮平均提取量为11.96±1.06 mg/g。
This research is about flavonoids extracted by ethanol reflux extraction method from peanut shells of Qinzhou city,Guangxi. The extractions are analyzed by spectrophotometry to determine the flavonoid content. Based on flavonoids extraction index,extracting experiments of different temperatures,time,liquid ratios and ethanol concentration were conducted. Besides,L9( 34) orthogonal experimental design is adopted to optimize the extraction conditions. The best extraction conditions are: extraction time 2. 5 h,ethanol concentration 70%,the ratio of liquid to solid 30 ∶ 1,extraction temperature 80 °C. Under these conditions,the average extraction amount of flavonoids was 11. 96±1. 06 mg / g.
