目的 探究肿瘤相关成纤维细胞(cancer-associated fibroblasts,CAF)对非小细胞肺癌(non-small cell lung cancer,NSCLC)生长的影响及可能机制。 方法 分离NSCLC患者CAF及对照正常成纤维细胞(normal fibroblasts, NF),体外观察CAF和NF对肿瘤细胞生长的影响,检测其分泌的细胞因子白介素-6 (interleukin-6,IL-6)的变化,MTT法检测IL-6对肿瘤细胞生长的影响;裸鼠接种人肺腺癌PC9细胞,同时接种CAF或NF,观察成纤维细胞对肿瘤生长具有影响。 结果 CAF能够在体外明显促进PC9细胞的增殖,CAF上清中IL-6表达明显增高,IL-6可促进肿瘤细胞的增殖;移植瘤模型进一步证实CAF对肿瘤生长的促进作用。 结论 CAF可通过高分泌IL-6促进NSCLC的增殖。
Objective To investigate the role of cancer-associated fibroblasts (CAF) in non-small cell lung cancer (NSCLC) and possible mechanisms. Methods Primary cultures of CAF and normal fibroblasts (NF) from NSCLC patients were established. The effect of CAF on the growth of NSCLC was observed. The expression of interleukin (IL)-6 and related molecules were detected by ELISA. The effect of IL-6 on the proliferation of NSCLC cells was evaluated by MTT assay. Results CAF had greater ability than NF to enhance the tumorigenicity of lung cancer PC9 cell lines. IL-6 was highly expressed in the CAF conditioned media, and further studies identified that IL-6 could promote the growth of NSCLC. CAF also can promote xenograft tumor growth in a mouse model. Conclusion CAF can promote the growth of NSCLC by secretion of IL-6.
