目的:建立环糊精手性固定相HPLC法分离艾迪康唑及其结构相似的抗真菌活性化合物。方法:采用Astec CycloBondⅠ2000DM环糊精手性柱,考察了环糊精固定相、流动相、pH值、缓冲液和柱温对拆分效果的影响。结果:确定最优化条件为固定相:Astec CycloBondⅠ2000系列环糊精手性柱(25cm×4.6mm,5μm),流动相:乙腈-0.1%乙酸三乙胺溶液(pH 6.0)=30∶70(V/V),流速:0.8ml/min;柱温:25℃;检测波长220nm;进样量:10μl。艾迪康唑及12种结构相似的抗真菌活性化合物的对映体均达到基线分离(Rs〉2.15)。结论:该方法简便快捷,可用于艾迪康唑及其结构相似的抗真菌活性化合物的手性拆分。
Objective:To establish a HPLC method using cyclodextrin-based chiral stationary phase for enantioseparation of antifungal drug iodiconazole and its structurally similar active compounds.Methods:Cyclodextrin chiral columns of Astec CycloBondⅠ 2000 series stationary phase were used.The effects of cyclodextrin stationary phase,mobile phase composition,pH value,buffer solution and column temperature on the separation of iodiconazole enantiomers were investigated.Results:The optimized conditions were as follows:the chiral stationary phase was Astec CycloBond Ⅰ 2000 DM column(25cm×4.6mm,5μm),the mobile phase was acetonitrile∶0.1%triethylamine acetate(30∶70,V/V)at a flow rate of 0.8ml/min,the column temperature was set at 25 ℃,the detection wavelength was 220 nm,and the injection volume was 10μl.Baseline separation of iodiconazole and its 12 structurally related active compounds were observed on the Astec CycloBondⅠ 2000 DM column(Rs〉2.15).Conclusion:This method is simple and rapid,and can be used for chiral separation of iodiconazole and its structurally similar antifungal compounds.
