MAWBP在急性溃疡性结肠炎模型中的作用

目的探讨MAWBP在溃疡性结肠炎急性模型中的作用。方法 40只BALB/c小鼠随机分成4组,每组10只,分别为正常对照组、Mock组、MAWBP组和MAWD组;Mock、MAWBP和MAWD组饮用4%DSS制作急性溃疡性结肠炎模型,正常对照组饮用高压过的饮用水。造模第1天,正常对照组、Mock组、MAWBP组和MAWD组分别注射慢病毒稀释液、慢病毒空载体、含MAWBP基因慢病毒液、含MAWD基因慢病毒液。造模期间进行疾病活动指数评分;造模结束检测各组小鼠肠道MAWBP蛋白和m RNA表达水平,病理切片评估肠道炎症程度。结果与正常小鼠相比,急性溃疡性结肠炎模型小鼠MAWBP蛋白表达水平明显降低;与Mock组相比,MAWBP组MAWBP蛋白和m RNA表达水平明显升高;MAWBP组小鼠疾病活动指数评分和低于Mock组;病理切片显示MAWBP组肠道炎症水平低于Mock组。结论 MAWBP对溃疡性结肠炎急性模型具有保护作用,可通过上调MAWBP表达改善溃疡性结肠炎病情。

英文摘要:

Object To study the effect of MAWBP on acute colitis model in mice.Methods Forty BALB/c mice were assigned randomly into four groups(Normal Control,Mock,MAWBP and MAWD group)with 10 mice per group.Mock,MAWBP and MAWD group drank 4% DSS solution while Normal Control group received autoclaved drinking water.On the first day of the model,Normal Control,Mock,MAWBP and MAWD group received lentivirus diluents,lentivirus vector,lentivirus with MAWBP gene and lentivirus with MAWD gene respectively.Meanwhile,Disease Activity Index was evaluated.At the end of the model,the protein and m RNA expression of MAWBP were detected and histological colitis severity were determined in colonic tissue.Results Mice with acute colitis had a lower protein expression of MAWBP than healthy mice.Mice in MAWBP group had a higher protein and m RNA expression of MAWBP than mice in Mock group.Mice in MAWBP group had a lower score of Disease Activity Index than mice in Mock group.Mice in MAWBP group developed a less severe inflammation than mice in Mock group.Conclusion MAWBP has a protective effect on acute colitis model in mice and up-regulation of MAWBP alleviates ulcerative colitis.
