目的探讨邻苯二甲酸二丁酯(DBP)对小鼠淋巴瘤细胞(EL4)的毒作用及维生素E(VitE)对细胞的保护作用。方法分别用10、100、500、1000μmol/LDBP作用细胞24h,并且VitE进行干预,检测细胞增殖、乳酸脱氢酶(LDH)活力及丙二醛(MDA)含量。结果DBP作用EL4细胞24h,100~1000μmol/LDBP均能明显促进EIA细胞增殖。1000μmol/LDBP可明显升高EL4细胞上清液中LDH活力及MDA含量。与1000μmol/LDBP组比较,50μmol/L VitE能显著降低细胞上清液中LDH活力及MDA含量。结论DBP使EL4细胞产生氧化损伤,这可能与DBP的免疫毒性有关。
Objective To investigate the cytotoxicity ofp-dibutyl phthalate (DBP) on EIA cell and the protective role of vitamin E (VitE). Methods EIA cells were exposed to different concentrations of 10, 100,500 and 1 000 μmol/L DBP respectively for 24 h, then received treatment with different concentrations of VitE; then to detect the status of cell proliferation, LDH activity in supernatant and MDA levels in cultural cells. Results The exposure to 100 - 1 000μmol/L DBP for 24 h, might promote the proliferation of EIA cell; 1 000 μmol/L DBP exposure might significantly increase the LDH activities in supernatant and MDA levels in cells. While 50 μmol/L VitE could significantly decrease the LDH activities and MDA levels . Conclusions DBP produces the oxidation injury in EIA cell, which may be related to its immunotoxicity.
