目的研究邻苯二甲酸二(2-乙基己基)酯(DEHP)对原代淋巴细胞分泌Th1、Th2型细胞因子的作用,进而探讨其对Th1/Th2平衡的影响。方法分别用刀豆蛋白(ConA)及佛波酯+离子霉素(PMA+Ion)去激活脾淋巴细胞,以不同浓度DEHP染毒细胞72 h及96 h,实验终点检测细胞增殖、细胞上清液中乳酸脱氢酶(LDH)活力及Th1型细胞因子[白细胞介素-2(IL-2)、γ干扰素(IFN-γ)]、Th2型细胞因子(IL-4、IL-6)的蛋白表达。结果10、50μmol/L DEHP单独作用细胞72 h明显促进细胞增殖。10、50μmol/L DEHP作用激活的淋巴细胞72 h显著增加了细胞上清液中LDH活力。50μmol/L DEHP作用淋巴细胞72 h能明显降低IL-2蛋白含量;1、10、50μmol/L DEHP 72 h则显著降低IFN-γ及IL-4含量。而DEHP对IL-6蛋白影响不显著。10、50μmol/L DEHP 96 h均明显降低了Th1/Th2比值。结论不同激活条件下,DEHP对淋巴细胞分泌细胞因子的影响不同,并能影响Th1/Th2平衡。
Objective To investigate the effect of di(2-ethylhexyl) phthalate(DEHP) on Th1 and Th2 type cytokines in primary lymphocytes,and then to study the effect of DEHP on Th1/Th2 balance.Methods ConA and(PMA+Ion) were respectively used to activate lymphocytes.Cells were exposed to different concentrations of DEHP for 72 hours and 96 hours.Cell proliferation and lactate dehydrogenase(LDH) activities were measured.Th1-type cytokines(IL-2,IFN-γ) and Th2 type cytokines(IL-4,IL-6) secreted in the cells were determined by ELISA after different treatments. Results DEHP at 10 and 50 μmol/L obviously increased cell proliferation and LDH activities in 72-hour treated cell supernatant under activated condition. DEHP at 50 μmol/L significantly reduced IL-2 content, and DEHP at 1,10,50 μmol/L obviouslydecreased contents of IFN-γ and IL-4 at the 72th hour. The effect of DEHP on IL-6 was not obviously. DEHP at 10 and 50 μmol/L interfered Th1/Th2 ratio at the 96th hour. Conclusion DEHP expresses different effects on Th1 and Th2 type cytokines under different activated conditions ,and disturbs Th1/Th2 balance.
