为研究三维点扩散函数层距与图像复原的关系,对数字共焦光学显微成像系统3D-PSF的构成、3D-PSF不同层距与图像复原效果的关系进行理论分析。通过仿真实验,研究了不同放大倍数的3D-PSF,在不同层距,相同层数和径向大小的情况下与图像复原效果的关系。实验表明:层距小于0.6μm时,放大倍数为40倍,图像在层距为0.5μm处取得最佳复原效果,放大倍数为20倍和10倍时,图像在层距为0.025μm处取得较好的复原效果。研究结果为在细胞切片的采集和三维显微图像复原实际应用中对3D-PSF的层距选取提供了理论依据和选取方法。
In order to study the relationship between the three dimensional point spread function and the image restoration, the structure of 3D-PSF and the relationship between the effect of different layer distance and image restoration of digital confocal optical microscope imaging system are ana- lyzed in theory. The relationship between the 3D-PSF of different distance and same layer number and radial amount is studied in different magnifications. Simulation results show that the best image restoration effect is obtained at the layer distance of 0. 5 μm when the distance is less than 0. 6μm and the magnification is 40 times. When the magnification is 20 times or 10 times, better image restoration is achieved at the layer distance of 0. 025μm. The research results provide a theoretical basis for the selection of 3 D-PSF layer distance in the practical application of the collection of cell sections and the practical application of three dimensional microscopic image restoration.
