目的:观察温和灸对慢性内脏痛敏大鼠外周(结肠)促肾上腺皮质激素释放因子(CRF)及其受体1(CRFRl)表达的影响。方法:采用乳鼠结直肠扩张(CRD)刺激建立慢性内脏痛敏大鼠模型,选取双侧天枢、上巨虚穴进行温和灸干预。以腹部撤回反射(AWR)评分作为检测大鼠内脏痛敏的行为学指标;采用免疫组织化学法、实时荧光定量PCR法分别观察CRF、CRFRl蛋白及其基因在大鼠结肠的表达及温和灸对其的影响。结果:在不同强度的CRD刺激下,与正常组相比,模型大鼠AWR评分升高(P〈0.01)。温和灸组干预后,大鼠AWR评分下降(P〈0.01)。模型组大鼠CRF、CRFR1蛋白及mRNA表达均高于正常组(P〈0.01),温和灸干预后,CRF、CRFR1蛋白及mRNA表达均降低(P〈0.01)。假灸组与模型组比较,差异无统计学意义。结论:温和灸降低大鼠内脏痛敏与其抑制外周(结肠)CRF—CRFR1信号途径有关。
Objective: To observe the influence of warming moxibustion (WM) in rats with chronic visceral hyperalgesia on the protein and mRNA expression of corticotropin-releasing factor (CRF) and corticotrophin releasing factor receptor 1 (CRFR1) in colon. Methods: The chronic visceral hyperalgesia rat models were established by colorectal distention. The rats were intervened by warming moxibustion in the points of Tianshu (ST25) and Shangjuxu (ST37). Abdominal withdrawal reflex (AWR) score was used as the behavior index to detect the visceral pain sensitivity in rats. Protein and mRNA expression of CRF and CRFR1 were detected by immunohistochemistry and real time-PCR, respectively in colon and the effect of warming moxibustion was observed. Results: At different levels of colorectal distention pressure, compared with the normal group, AWR score increased in model group (P〈0.01). After the warming moxibustion intervention, the AWR score decreased (P〈0.01). The protein and mRNA expressions of CRF and CRFR1 in model group were higher than in normal group (P〈0.01), and after the intervention, the expressions decreased (P〈0.01). Compared the pretend moxibustion group with the model group, there was no significantly difference. Conclusion: WM treatment can attenuate CRD-induced chronic visceral hyperalgesia in rats and this effect might be due to WM reduction of colonic CRF-CRFR1 system.
