目的:考察TFU通过缓慢释放5-FU抑制人肝癌细胞生长,分析TFU代谢转化特点。方法:以96孔板接种人肝癌细胞SMMC-7721,加入不同剂量TFU,每隔24h取培养板,取细胞培养液,以HPLC法检测TFU和5-FU水平,并以MTT法测肿瘤细胞生存率。培养板加入肝微粒体酶,重复上述检测HPLC方法和MTT实验,比较加入肝微粒体酶前后培养液中TFU和5-FU水平,分析TFU通过缓慢释放5-FU抑制人肝癌细胞生长。结果:TFU对人肝癌细胞SMMC-7721抑制作用弱,加入肝微粒体酶后抑制作用增强。HPLC检测上清液中TFU和5-FU水平发现,TFU可能通过肝微粒体酶的介导作用,缓慢释放出5-FU并维持较长时间药物水平,这种代谢释放特点与TFU对肿瘤细胞生长抑制率有密切相关性。结论:TFU可能存肝微粒体酶介导下,通过缓慢水解方式释放出5-FU,维持对肝癌细胞较长时间的抑制生长作用。
Objective: We aimed to evaluate the efficacy of TFU as a precursor of 5 - FU on the growth inhibition of human hepatoeellular carcinoma cell line SMMC -7721. Methods: Human hepatoeellular carcinoma cells were seeded in 96 - well plate exposure to different concentrations of TFU without or with liver microsomal enzymes. The levels of TFU and 5 - FU in medium were analyzed by HPLC every 24 hours for five consecutive days. And then the cell viability was measured using MTT assay. The inhibition of cell growth was compared and the sustaining release of 5 -FU was analyzed. Result: TFU weakly prevented SMMC -7721 cell proliferation in the absence of liver mierosomal enzymes. Proliferation of SMMC -7721 cells was drastically suppressed in the presence of liver microsomal enzymes. HPLC analysis showed that TFU might transform into 5 - FU by hydrolyzing in the presence of liver microsomal enzymes. The cell growth inhibition of SMMC - 7721 may related to the sustaining release of 5 - FU from TFU. Conclusion TFU may inhibit the growth of human hepatocellular carcinoma cell via sustaining release of 5 - fluorouracil.
