目的研究上皮细胞间质转型(EMT)相关分子金属硫蛋白(MT)、低氧诱导因子-1α(HIF-1α)和锌指转录因子Snail在人甲状腺乳头状癌中的表达及其意义。方法用免疫组化SP法,检测79例甲状腺乳头状癌及12例结节性甲状腺肿中MT、HIF-1α及Snail的表达情况。结果79例甲状腺乳头状癌中,MT、HIF-1d和Snail的阳性率分别为56.96%,92.41%,53.16%,且三者的阳性表达对于TNM分期及淋巴结转移均具有显著性差异(P〈0.05)。MT与Snail的表达呈显著正相关(P=0.0004),MT与HIF-1α呈显著正相关(P=0.0450),Snail与H1F-1α呈显著正相关(P=0.0008)。结论推测MT、HIF-1α和Snail是针对甲状腺乳头状癌侵袭转移的早期发现及其预后评估的潜在分子标记物。
Objective To investigate the significance of metallothionein ( MT), hypoxia - inducible factor - 1α ( HIF - 1 α ) and Snail expression in patients with papillary thyroid carcinoma. Methods The expression of MT, HIF - 1α and Snail were analyzed with immunochemistry in 79 papillary thyroid carcinoma and 12 nodular goiters. Results Positive expression of papillary thyroid carcinoma in 79 cases of MT, HIF - 1α and Snail was shown in 56.96%, 92. 41% and 53.16% , respectively. Positive expression of MT, HIF - 1α and Snail in papillary thyroid carcinoma was associated with lymph -node metastasis as well as TNM stage. Positive expression of MT was significantly correlated with that of Snail ( P = 0. 0004 ) and that of HIF - 1 α(P = 0. 045 ) , while a significant correlation was showed between HIF - 1 α and Snail ( P = 0. 0008 ). Conclusion Coexpression of MT, HIF- 1α and Snail suggests not only an early marker of tumor cells metastasis, but also a significant prognostic predictor in patient with PTC.
