目的:观察补脾益气方对脾虚哮喘大鼠模型肺组织Toll样受体4(TLR4)表达的影响,探讨脾虚对哮喘大鼠气道炎症影响机制及补脾益气方对其的调控作用。方法:36只健康雄性Wistar大鼠,随机分为3组:对照组、脾虚哮喘组、脾虚哮喘治疗组,每组12只。采用RT-PCR检测大鼠肺组织TLR4 mRNA表达水平;采用免疫组化检测大鼠肺组织TLR4的蛋白表达水平。结果:和对照组比较,脾虚哮喘组大鼠肺组织TLR4 mRNA和蛋白表达水平显著增高(P〈0.01);和脾虚哮喘组比较,脾虚哮喘治疗组大鼠肺组织TLR4 mRNA和蛋白表达水平显著降低(P〈0.01)。结论:脾虚哮喘组大鼠肺组织中TLR4 mRNA及蛋白表达水平显著增高,补脾益气方能够降低脾虚哮喘大鼠肺组织中TLR4 mRNA和蛋白表达水平。
Objective: To observe the influence of Bupi Yiqi Formula on TLR4 expression in lung tissue of rats with spleen-dei cency bronchial asthma, and to discuss the inl uence of inl ammation mechanism of rats air passage with asthma caused by spleen-deficiency and the regulating effect of Bupi Yiqi Formula. Methods: 36 healthy male Wistar rats were randomly divided into 3 groups: control group, model group, Bupi Yiqi group, 12 rats in each group. Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of TLR4 mRNA in the tissue of the lung. Immunohistochemistry was used to test the expression of TLR4 protein. Results: Compared with the control group, the expression of TLR4 mRNA and protein in the lung tissue of rats in the model group was signii cantly higher(P〈0.01). Compared with model group, the expression of TLR4 mRNA and protein in the lung tissue of rats in Bupi Yiqi group was signii cantly lower(P〈0.01). Conclusion: The expression of TLR4 mRNA and protein in the lung tissue of rats in the model group increases signii cantly, and the expression level of TLR4 mRNA and protein in the lung tissue of rats with asthma can be decreased by Bupi Yiqi Formula.
