目的观察培土生金法对脾虚哮喘模型大鼠TNF—α/NF—κB信号通路变化的影响,探讨培土生金法干预哮喘气道炎症和黏液高分泌的信号转导机制。方法健康雄性Wistar大鼠50只,随机分成5组:对照组、脾虚哮喘组、哮喘组、脾虚哮喘治疗组和哮喘治疗组,每组10只。采用ELISA试剂盒法测定血清和BALF中TNF-α的含量,采用免疫组化测定大鼠肺组织NF—κBp65的表达和入核率。结果脾虚哮喘组和哮喘组血清和BALF中TNF—α含量及大鼠支气管上皮细胞中NF—κBp65阳性表达均显著升高,和对照组比较差异显著(P〈0.01);但是脾虚哮喘组和哮喘组比较,血清和BALF中TNF-α含量无显著性差异。与哮喘组比较,脾虚哮喘组大鼠支气管上皮细胞中NF—κB-p65入核率显著增加(P〈0.01)。和脾虚哮喘组比较,脾虚哮喘治疗组血清和BALF中TNF—α含量及肺组织NF—κBp65入核率显著降低(P〈0.01);和哮喘组比较,哮喘治疗组血清和BALF中TNF—α含量及肺组织NF—KBp65入核率显著降低(P〈0.01)。结论培土生金法能够降低脾虚哮喘和哮喘大鼠血清和BALF中TNF—α的含量和肺组织NF—κBp65入核率。
Objective To observe the effect of reinforcing earth to strengthen metal on TNF-α/NF-KB signa- ling pathway diversification in rats spleen-deficency in bronchial asthma , the relationship of airway mucus hypersecretion formation signal transduction mechanism and the effective mechanism of reinforcing earth to strengthen metal were explored. Methods 50 healthy male Wistar rats were randomly divided into 5 groups: control group, spleen deficiency asthma group, asthma group, spleen deficiency asthma treatment group, asth- ma treatment group, 10 rats in each group. Concentration of TNF-α in serum and BALF was detected by ELISA, and expression and nucleation translocation ratio of NF-KBp65 was detected by immunohistochemis- try too. Results TNF-α levels in BALF and NF-KBp65 expression in bronchial epithelial cells of rat were significantly increased in spleen deficiency asthma group and asthma group. Compared with the control group, spleen deficency asthma group and asthma group had significant difference (P 〈 0. 01 ). TNF-α in se- rum and BALF showed no significant difference between spleen deficiency asthma group and asthma group. Compared with asthma group, into nucleation rate of NF-KBp65 in rat bronchial epithelial cells of spleen de- ficiency asthma group increased significantly (P 〈 0. 01 ). Compared with spleen deficiency asthma group, TNF-α in serum or BALF and into nucleation rate of NF-KBp65 significantly reduced in spleen deficiency asthma treatment group (P 〈 0. 01 ). Compared with asthma group, TNF-α in serum or BALF and into nucle- ation rate of NF-KBp65 significantly reduced in asthma treat-ment group( P 〈 0.01 ). Conclusions TNF-α in serum or BALF in spleen deficency asthma or asthma rat and into nucleation rate of NF-KBp65 in spleen deficency asthma or asthma rat's lung tissue can be significantly reduced by the effect of reinforcing earth to strengthen metal.
