目的:预测结核分枝杆菌(MTB)相关抗原Rv1258c、Rv1410c及Rv3425的HLA限制性细胞毒性T细胞(CTL)表位。方法:通过NCBI数据库获取各蛋白的氨基酸序列,利用BLAST分析其与人类蛋白质的同源性,利用SYFPEITHI、BIMAS和NetCTL数据库预测分析MTB相关抗原的HLA-A2与HLA-A3限制性CTL表位。结果:初步筛选出3个候选抗原的HLA-A2和HLA-A3限制性CTL优势表位,分别为Rv1258c343~351、380~388、290~298、52~60;Rv1410c510~518、470~478、452~460、420~428;Rv3425118~126、43~51、29~37。结论:预测出MTB3个相关抗原的HLA-A2与HLA-A3限制性CTL表位,为抗结核多表位疫苗的研究奠定了基础。
Aim:To predict the HLA restricted cytotoxic T lymphocyte(CTL)epitopes in Mycobacterium tuberculosis antigens Rv1258c,Rv1410c and Rv3425.Methods:The amino acid sequences of the antigens were obtained from NCBI database.Their homologies to human proteins were analyzed by BLAST.HLA-A2-and HLA-A3-restricted CTL epitopes were predicted by SYFPEITHI,BIMAS and NetCTL.Results:The predominant HLA-A2-and HLA-A3-restricted CTL epitopes predicted from the three candidate antigens were Rv1258c 343~351,380~388,290~298,52~60;Rv1410c 510~518,470~478,452~460,420~428;and Rv3425 118~126,43~51,29~37.Conclusion:Eleven HLA-A2-and HLA-A3-restricted CTL epitopes have been predicted from Mycobacterium tuberculosis antigens Rv1258c/Rv3425/Rv1410c,which could be used in research of antituberculosis multi-epitope vaccine.
