目的:采用简易微列阵法检测谷氨酸递质系统中与精神分裂症相关联的基因。方法:采用简易微阵列法,即PCR-等位基因特异性引物延伸和硝酸纤维素膜杂交法分析云南省255例精神分裂症患者和262名对照4个候选基因的4个SNP位点[rs778293(G72/G30)、rs2299225(GRM3)、rs11146020(GRIN1)和rs2814351(GRID1)]与精神分裂症的关联性。结果:对照组谷氨酸递质系统中的4个基因的4个SNP位点均符合Hardy-Weinberg遗传平衡定律。4个位点中,rs778293位点与精神分裂症存在关联性(基因型频率χ2=14.017,等位基因频率χ2=14.951,P〈0.05)。结论:G72/G30基因SNP多态性与精神分裂症有关。
Aim:To identify the associated genes in patients with schizophrenia from Yunnan province.Methods:The DNA samples of 255 schizophrenic patients and 262 controls from Yunnan province were collected.Four single nucleotide polymorphism(SNP) sites of four candidate genes in glutamic acid neurotransmitter system [rs778293(G72/G30),rs2299225(GRM3),rs11146020(GRIN1),and rs2814351(GRID1)] were analyzed by using PCR,allele-specific primer extension,and hybridization in nitrocellulose membrane,respectively.Results:The four SNP sites in the four candidate genes of the control met the Hardy-Weinberg law.Among the 4 SNPs sites,rs778293 was associated with schizophrenia(genotype frequency:χ2=14.017;allele frequency:χ2=14.951,P0.05).Conclusion:The SNP of G72/G30 gene may be associated with schizophrenia.
