建立了高效液相色谱法测定3个唑来膦酸衍生物——1-羟基-3-(1£卜咪唑-1-基)丙烷-1,1-双膦酸、1-羟基-4-(1H咪唑-1-基)丁烷-1,1-双膦酸和1-羟基·5-(1胁咪唑-1-基)戊烷-1,1-双膦酸。使用C.8色谱柱,以4mmol/L磷酸二氢钠缓冲液(pH5.3)-甲醇(30:70,含5mmol/L溴化四丁铵)为流动相,检测波长210nm。上述3个化合物在0.2~lmg/ml范围内线性关系良好。回收率分别为101.7%、100-3%和99.8%,RSD为1.26%、1.43%和1.61%。
An HPLC method was established for the determination of three zoledronic acid derivatives: 1 -hydroxy-3- (1H-imidazol- 1-yl) propane- 1,1-diphosphonic acid, 1-hydroxy-4- (1H-imidazol- 1 -yl) butane- 1,1-di- phosphonic acid and 1-hydroxy-5-(1H-imidazol-l-yl)pentane-l,l-diphosphonic acid. A C18 column was used with the mobile phase of 4 mmol/L disodium hydrogen phosphate buffer (pH 5.3)-methanol [30 : 70, containing 5 mmol/L tetrabutyl ammonium bromide (TBAB)] at the detection wavelength of 210 nm. The calibration curves were linear in the range of 0.2 - 1 mg/ml. Their recoveries were 101.7%, 100.3 % and 99.8%, with RSDs of 1.26%, 1.43 % and 1.61%, respectively.
