目的:对比研究连翘提取物对不同致炎物大鼠足肿胀的作用,探讨连翘提取物的抗炎机理。方法:通过建立角叉菜胶和新鲜蛋清致大鼠足肿胀模型,在造模前0.5 h分别给大鼠灌胃给药10 m L/kg 0.5%吐温80、0.2 g/kg阿司匹林、5 g/kg连翘提取物,测量造模后0、0.5、1、2、3、4 h大鼠足趾体积。结果:致炎各组大鼠足趾后0.5 h均开始肿胀,连翘提取物可减轻蛋清致足肿胀程度,但对角叉菜胶致肿胀模型的抑制作用比蛋清致足肿胀模型强,延缓足肿胀,在2 h抑制作用最强。结论:研究结果表明连翘提取物对角叉菜胶致大鼠足肿胀模型有一定抗炎作用,其机理可能与促进COX-2产生,抑制PGE_2、PGD_2、6-keto-PGF1α和TXB2的合成有关。
Objective: To compare the anti-inflammation effect of Forsythia extract on different paw edema rats and explore the anti-inflammation mechanism. Methods: Rat paw edema models were established by carrageenan( 0. 1 m L/paw) and fresh egg white( 0. 1 m L/paw) in the study. 0. 5 h before establishing models,rats were orally administered with 0. 5% Tween-80( 10 m L /kg),aspirin( 0. 2 g/kg) and Forsythia extract( 5 g/kg) respectively. And then,the rats paw volume was measured at 0,0. 5,1,2,3,4 h after model establishment. Results: The results showed that both groups paw began to swell at 0. 5 h. Forsythia extract reduced egg white-induced paw edema and the inhibition effect on carrageenan-induced paw edema was stronger than which on egg white-induced paw edema model. Conclusion: Forsythia extract could retard the progress of swelling and process the higher inhibition effect at 2 h. In conclusion,these findings suggest that Forsythia extract possess anti-inflammation activity on carrageenan-induced paw edema rats. It is possibly related to the mechanism of promoting COX-2 production and inhibiting synthesis of PGE_2,PGD_2,6-keto-PGF1α and TXB_2.
