以植物乳杆菌(Lactobacillus plantanum) AB-1为目标菌株,考查了培养基中亚硝酸钠(NaNO2)含量、初始pH、培养温度、接种量及培养时间对其降解NaNO2的影响.在此基础上,利用响应面试验分析了培养温度、接种量和培养时间的交互作用对该菌株降解NaNO2的影响.结果表明,培养温度、接种量及培养时间对菌株AB-1降解NaNO2具有明显影响,建立的响应面方程可以实现对该菌株降解NaNO2条件的优化.验证试验结果显示,当MRS培养基中NaNO2含量为150 mg/L、初始pH值为6.5、培养温度为32℃、接种量为2.7%,培养时间为28h时,植物乳杆菌AB-1对NaNO2的降解率为97.94%,与预测值(98.19%)几乎一致.
In this study,a strain of Lactobacillus plantmun AB-1 was used to degrade NaNO2 in medium.The effect of initial NaNO2 content,initial pH,cultivation temperature,inoculum and cultivation time on the degradation of sodium nitrite were investigated.Moreover,the interactions of each factor between inoculum,cultivation temperature and time were analyzed by response surface methodology.The results showed that inoculum,cultivation temperature and time showed significant effect on NaNO2 degradation,and the optimum NaNO2 degradation condition was obtained through the establishment of response surface equation.Verification results showed that optimal condition was NaNO2 content 150 mg/L in MRS medium,initial pH 6.5,inoculum 2.7%,cultivation temperature 32 ℃ and time 28 h.Under this condition,verification test proved that NaNO2 degradation rate was 97.94%,which was almost the same as the predicted value (98.19%).
