初步分离纯化了链霉菌JD211所产抗菌活性物质。结果显示:活性产物最易溶解在正丁醇中,其次为乙酸乙酯;微溶于氯仿;不溶于苯、乙醚和石油醚等低极性的有机溶剂。硅胶柱层析中用1∶5的石油醚:乙酸乙酯作为洗脱剂时的洗脱液具有最大的抑菌活性。薄层层析实验结果表明:以1∶5的石油醚:乙酸乙酯作为展开剂展开时,Rf值为0.53和0.6时具有抑菌活性。液相色谱-质谱联用分析表明活性物质与放线菌酮标准品具有相同的分子量,都为281.25。确定活性物质为戊二酰亚胺类抗生素。
The isolation,purification and identification of the antifungal active substances from Streptomyces JD211 were conducted.The results showed that the antifungal active substances were most inclined to dissolve in n-butanol,followed by ethyl acetate;slightly soluble in chloroform;insoluble in benzene,ether and petroleum ether and other low-polar organic solvents.Column chromatography on silica gel using 1 ∶ 5 petroleum ether:ethyl acetate as the eluent agent had the greatest inhibitory activity.TLC results showed that 1 ∶ 5 petroleum ether:ethyl acetate which were used as the eluent to start,when Rf values were 0.53 and 0.6,had antibacterial activity.Liquid chromatography-mass spectrometry analysis showed that the active substance and actidione standard had the same molecular weight,both were 281.25.The active substance was determined as glutarimide antibiotics.
