目的探讨以滋阴益气、清热利湿、化痰消瘀组方的左归降糖清肝方对高脂饮食2型糖尿病(T2DM)转基因MKR小鼠脂肪肝发生中脂代谢相关基因mRNA表达的影响。方法30只MKR小鼠随机分为MKR组、MKR高脂组和左归降糖清肝方组,左归降糖清肝方干预治疗高脂饮食诱发脂肪肝的MKR鼠4w;观察各组小鼠肝脏形态,RT-PCR检Nd,鼠肝脏脂代谢相关基因mRNA表达变化。结果①MKR高脂组较MKR组小鼠肝脏乙酰辅酶A羧化酶(ACC-α)、脂肪酸合酶(FAS)和内质网甘露糖苷酶样蛋白1(EDEM1)的mRNA表达增加,apoB100mRNA表达降低;②左归降糖清肝方降低高脂饮食MKR小鼠肝脏ACC-α、FAS和EDEMI的mRNA表达,增加apoB100mRNA的表达。结论左归降糖清肝方能通过调节肝脏脂代谢相关基因mRNA表达,缓解肝脏脂肪性变。
Objective To observe the effects of Zuogui Jiangtang Qinggan recipe(ZJQR) on expression of lipid metabolism related gene in MKR mice of type 2 diabetes combined with liver steatosis raised by high fat feeds. Methods Thirty MKR mice were randomly di-vided into the control group(CG) , the model group(MG) and the ZJQR group(ZG). The ZG mice were raised by high fat feeds to form liver steatosis and had been administered by ZJQR for 4 weeks then. Pathological changes of the liver were examined, and RT-PCR was used to analyze the related gene expressions of lipid metabolism in the liver of MKR mice. Results ①The mRNA expressions of ACC-α, FAS and EDEM1 in MG mice were elevated significantly compared with CG, and the mRNA expression of apoB100 was decreased; ②ZJQR could de-crease the gene expressions of ACC-α, FAS and EDEM1, whereas increase the gene expression of apoB100 in MKR mice raised by high fat feeds. Conclusions ZJQR can relieve the liver steatosis through regulating the related gene expressions of lipid metabolism in the liver.
