利用静电吸附作用将抗体羊抗人免疫球蛋白G(anti-HIgG)修饰在银电极表面,制备了无标记型的免疫电极,采用电化学方法对人免疫球蛋白G(HIgG)进行分析测定。在0.1mol/L K3PO4-K2HPO4(pH 7.2)缓冲液中,加入的HIgG与银电极表面的anti-HIgG发生免疫反应,使anti-HIgG的电荷减少产生脱附,裸露出银,使电极有效面积增大,银溶出电流发生改变。利用电流变化和HIgG浓度之间的关系,实现对HIgG的分析测定,电流的变化量与HIgG的含量在0~140ng/mL范围内成正比关系。该法具有灵敏度高、无须标记,使用方便等优点。
A label-free electrochemical immunoassay was proposed for human immunoglobulin G (HIgG) determination. Anti-HIgG was adsorbed on a silver electrode by electrostatic interaction to obtain an immuno-electrode,and the surface of the silver electrode was blocked by Bovine serum albumin. An insula- tion layer was formed on the electrode surface. In 0.1 mol/L K3PO4-K2HPO4 (pH 7.2) buffer solution, the immunoreaetion between HIgG and anti-HIgG occurred and made the charge of the adsorbed anti-HIgG changed. The anti-HIgG covered on silver electrode surface desorbed and led to the silver surface exposure to solution. The effective area of silver electrode increased which caused the stripping current of silver to rise up. The anodic stripping current of silver was proportional to the concentration of HIgG in the range of 0 - 140 ng/mL, which presented a stripping voltammetric immunoassay for HIgG determination. This method showed the characterization of high sensitivity, label-free and easy operation.
