[目的]采用样本熵分析法和频率分析法分析电针“足三里”穴对孤束核神经元兴奋性的影响,探讨两种分析方案分析针灸干预效应的可行性和差异。[方法]雄性健康SD大鼠10只,随机分为空白组和电针组,每组5只,均行颅内微丝阵列电极植入术。电针组双侧“足三里”穴行一次电针刺激,参数如下:连续波,2/100Hz,1-2mA,刺激30min,空白组仅予相同固定。记录电针前、后各10min孤束核神经元放电。行频率分析,根据神经元放电频率改变,将神经元分为兴奋性、抑制性和无反应三类,并计算其分布和放电频率变化率,对峰电位间隔行样本熵分析,最后对两者行相关性分析。[结果]电针后,电针组大鼠孤束核神经元三类神经元分布与放电频率,与对照组相比无明显差异( P>0.05);电针前,对照组和模型组样本熵无明显差异( P>0.05);电针后,与对照组相比,电针组大鼠孤束核峰电位间隔样本熵明显降低( P<0.05)。相关性分析提示,电针后大鼠孤束核放电频率改变与峰电位间隔样本熵具有负相关性( P<0.05)。[结论]样本熵和放电频率均可用于电针对神经兴奋性干预作用的分析,频率分析易于掌握,可用于删选和初步分析,样本熵适用于深入分析,但方法复杂,需要分析人员有一定数学基础。
[Objective] Two analysis methods, spike frequency and inter spike interval(ISI) Sample Entropy(SampEn), were used for investigating the regulatory effect of electroacupuncture(EA) at “ Zusanli” on nucleus of the solitary tract(NTS), in order to discuss their feasibility and difference when used in EA analysis. [Methods] 10 rats were randomly divided into control and EA groups. All rats underwent electrode implantation surgery. Rats in EA groups received once treatment with EA(constant aquare wave, 2/100Hz frequencies, intensities ranging from 1~2mA), and the NTS neuronal activations were recorded before and after EA. The frequency of spike and the SampeEn of ISI were analyzed in all rats. [Results] Compared with control group, EA did not regulate the change rates of frequency and population of different types of NTS neurons(P〉0.05). However, EA decreased the SampEn of EA group at 10min after EA(P〈0.05). [Conclusion] Both of the two analysis methods could be used investigating the effect of acupuncture. Frequency could be used in primary analysis and the SampEn analysis could be used in deep analysis.
