运用单因素和正交试验设计对洋虫超氧化物歧化酶(SOD)的提取工艺进行了优化研究,得到最优的提取条件为:用m(洋虫粉末):V(抽提液)=1g:13mL的0.13mol/L NaCl溶液在28℃下抽提14h。活性可达38.83U/mL,比活力(以蛋白计)可达123.66U/mg。结果表明,NaCl溶液比传统的磷酸盐缓冲液抽提效果更好,其抽提浓度对SOD的提取结果影响最大;同时得到SOD的最佳热变性温度为60℃,时间为15min。
The extraction conditions of superoxide dismutase (SOD) from Martianus dermestoides was optimized by single factor trial and orthogonal experimental design. The optimal extraction conditions for SOD from M. dermestoides were obtained as extraction solution 0. 13 mol/L NaCl, ratio ofM. dermestoides powder to extraction solution 1 g: 13 mL, extraction temperature 28 degrees C, and extraction time 14 hours. The activity of SOD was 38.83 U/mL, and the specific enzymatic activity of SOD was 123.66 U/mg under the optimal extraction conditions. Result showed that SOD extracted by NaCl had a higher activity compared with the conventional phosphate extraction method. The concentration of extraction solution had the greatest effect on SOD extraction. During the extraction process, most of the unwanted protein could be removed by a heat denaturation at 60 degrees C for 15 minutes.
