利用SRAP技术,从72对引物中筛选出10对引物对帽儿山地区的10种不同色斑型(2种斑型组)异色瓢虫进行扩增,对扩增结果进行基因组多态性分析,结果显示,10对引物共扩增出条带175条,其中多态性条带为133条,多态位点比率为76.04%。利用POPGENE32软件进行不同斑型组遗传多样性指数的分析,通过非加权配对算术平均法(UPGMA)进行聚类分析,结果表明,帽儿山地区异色瓢虫的遗传多样性较为丰富,部分异色瓢虫的亲缘关系与其底色之间或者色斑形状之间存在一定的关联性,与形态分类结果一致。
Ten pairs of primers screened from seventy-two pairs were used to amplify the whole genome of ten splash types of Hannonia axyridis Pallas, which were collected from Maoershan region of Shangzhi County, Heilongjiang Province, by the Sequence-related Amplified Polymorphism (SRAP) technique. The genomic polymorphism of this amplification result was analysed. A total of 175 bands were generated, in which 133 bands were polymorphie bands and the percentage of polymorphie loci was 76.04 percent. The genetic diversity of different splash types was analysed using POPGENE32. Unweighted pair group method with arithmetic mean (UPGMA) was used in cluster analysis. Results indicate that there is a high genetic diversity within H. axyridis. There is a certain correlation between affiliation and ground colour or between splash types for some H. axyridis, which accord with the results of morphological taxonomy.
