通过磁珠富集法首次筛选黄颡鱼微卫星标记并对其特征进行分析。用生物素标记的寡核苷酸探针(AC)8、(CT)8、(AT)7、(GATA)8和(GATT)7进行微卫星片段的富集,挑选部分阳性克隆测序获得38个含有微卫星的DNA序列(GenBank登录号:EF473162-EF473200),设计并合成了22对微卫星引物。以普通黄颡鱼(Pseudobagrus fulvidraco)基因组DNA为模板,进行PCR扩增,结果筛选出18个多态性微卫星标记,每对引物等位基因数2-8个(平均3.78),遗传杂合度0.2225-0.8101(平均0.5755),多态信息含量0.3425-0.7900(平均0.5336)。此外18对多态性微卫星引物中有16对在黄颡鱼属其它4种鱼类具有通用性,13对在鲇形目(Siluriformes)中的3种鱼类具有通用性。因此,可以推断筛选的多态性微卫星标记可用于黄颡鱼属和鲇形目鱼类的遗传分析。
Isolation and characterization of microsatellites from Pseudobagrus fulvidraco were carried out using an enrichment protocol. The size-fraetionated genomic library was screened with (AC)8, (CT)8, (AT)7, (GATA)8 and (GATT)7 oligonucleotide probes, respectively. A total of 38 clones(GenBank No. EF473162-EF473200) were identified and 22 primer pairs with microsatellite repeats were designed. Of the 22 primer pairs, 18 amplified polymorphic loci in P. fulvidraco. Among the polymorphic loci, the number of alleles per locus varied from 2 to 8 (average 3.78 ), the observed heterozygosities ranged from 0.2225 to 0.8101(average 0.5755) and the polymorphism information content were from 0.3425 to 0.7900(average 0.5336). These 18 primer pairs were also used to investigatefor Pseudobagrus and Siluriformes species. Sixteen of the 18 primer pairs amplified the loci in the Pseudobagrus species, and 13 pairs amplified the loci in the Siluriformes species. These results indicated that the microsatellites isolated in P.fulvidraco were highly informative and could be useful for genetic analysis in the Pseudobagrus and the Siluriformes species.
