免疫共抑制分子PD-1为CD28超家族成员,其最初是从凋亡的小鼠T细胞杂交瘤2B4.11克隆出来。PD-1基因敲除小鼠可诱导多种自身免疫性疾病,表明PD-1是一种重要的免疫抑制分子并在外周耐受中发挥着重要的作用。因此,以PD-1为靶点的免疫调节对抗肿瘤、抗感染、抗自身免疫性疾病及器官移植存活等均有重要的意义。
PD-1,firstly cloned from mouse T cell hybridoma 2B4.11,belongs to members of CD28 superfamily.Some autoimmune diseases would be developed from PD-1 gene knocked out mice,which indicating that PD-1 is a co-inhibitory molecule which actively participates in maintaining peripheral tolerance.So,PD-1 would be a useful target for the immunologic therapy of carcinoma,infection,autoimmune diseases as well as organ transplantation in the near future.
